Effects of a-keto-6-guanidinovaleric acid on inhibitory amino acid responses on mouse neurons in cell culture

چکیده
The experimentally proven convulsant a-keto-6-guamdinovaleric acid (a-K-6-GVA) was applied to mouse spinal cord neurons in primary dissociated cell culture to assess its effects on postsynaptic y-aminobutync acid (GABA)and glycine (GLY)-responses. Intracelltaar microelectrode recording techniques were used. a-K-b-GVA reversibly inhtbited both GABAand GLY-respones m a concentration-dependent manner, The effect of a-K-6-GVA on GABA-responses was not antagomzed by co-application of the benzod~azepine receptor antagonist CGS 9896 The results suggest that a-K-6-GVA inhibited responses to the inhibttory neurotransmitters GABA and GLY by blocking the chloride channel. This action might underlie the convulsant effect of this compound in rabbit. The possible pathophysiological importance of a-K-t~-GVA in hyperargmlnemlc patients ~s discussed.

Guanidino compounds that are increased in hyperargininemia inhibit GABA and glycine responses on mouse neurons in cell culture.
The effects of arginine, homoarginine, alpha-keto-delta-guanidinovaleric acid and argininic acid (guanidino compounds that were found to be increased in hyperargininemia) were evaluated on responses to gamma-aminoburtyric acid (GABA) and glycine (Gly) on mouse neurons in primary dissociated cell culture. GABA and Gly were applied iontophoretically and intracellular microelectrode recording tech...

Introduction: Embryonic Stem (ES) cells as pluripotent cells derived from the inner cell mass of blastula can differentiate to neural cells in vitro and this property is valuable in studies of neurogenesis and in the generation of donor cells for transplantation. In this regard, the propose of this research, was the study of the role of two important factors in the development of neural syst...

Four guanidino compounds that have been found to be markedly increased in cerebrospinal fluid and brain tissue of uremic patients, namely, guanidine, methylguanidine, creatinine, and guanidinosuccinic acid, were applied to mouse spinal cord neurons in primary dissociated cell culture to evaluate their effects on postsynaptic responses to gamma-aminobutyric acid (GABA) and glycine. Intracellular...

Lysophosphatidic acid (LPA) known as a serum-derived growth factor, is involved in several cell physiological functions in the female reproductive system including: oocyte maturation, in vitro fertilization and embryo implantation by its transmembrane G protein-coupled receptors. The aim of the present study was to examine the effect of LPA on in vitro follicular development o...
